SENSITIVITY TESTING OF KASTLE MEYER AND LEUCOMALACHITE GREEN AS PRESUMPTIVE TESTS FOR DETECTION OF DRIED BLOOD

Abstract

To characterize the prevalence of blood contamination on glucose strips used in hospitals, a first of its kind study is in need to be conducted which will recommend monitoring actions to help decrease the risk of spread of infectious agents within hospital environment.  Each glucose strip out of almost 20 vials, each containing 20 glucose strips, will be inspected visually and sampled for testing with Kastle Meyer test and Leucomalachite Green test for hemoglobin to detect the presence of blood. A preliminary study for validating the tests and testing the sensitivity of both these tests was conducted. The sensitivity was determined by testing serial dilutions of whole blood samples in the series of 10-1, 10-2, 10-3, 10-4, 10-5 and 10-6 using 0.9% normal saline as diluents. The whole blood and normal saline were used as positive and negative control for the test. Triplicate experiment was performed on each of the dilutions to validate the two tests. Findings of this study indicate that the Kastle Meyer test is relatively most sensitive.
