INCREASING BIOMASS ACCUMULATION AND WITHAFERIN A PRODUCTION IN CELL SUSPENSION CULTURE OF WITHANIA SOMNIFERA

Withania somnifera, commonly known as Ashwagandha, is a valued herb in Ayurvedic medicine. Roots, leaves and preparations of the plant are traditionally used as tonic, hypnotic, sedative and diuretic. W. somnifera mainly contains withanolides which are biologically active secondary metabolites present in roots and leaves. In the present study, we have standardized the protocol for the extraction and purification of Withaferin A from the cell suspension cultures of W. somnifera. Withanferin A is having a high medicinal value and possesses potent anti-tumor and antioxidant properties. In the present study the callus was obtained from leaves as explant. Highly profuse and friable callus was then used as inoculum for suspension culture. Here different combinations of phytohormones viz. IAA+BAP (1.0+0.8mg/l) and NAA+ BAP (0.5+1mg/l) were used. This study involves a detailed investigation about the accumulation of secondary metabolite content, in in vivo cell suspension, both quantitatively and qualitatively. Pure compounds were extracted and identified using the simple techniques viz., Thin Layer Chromatograpy and High Pressure Liquid Chromatography. The growth of biomass in suspension culture of W. somnifera was calculated in interval of subsequent culture days in terms of fresh biomass of callus. After crossing lag phase, the cells in suspension culture entered log phase and maximum increase in biomass was observed between 15-20 days. Thus the maximum callus biomass 0.41g/80ml was observed in suspension culture from NAA+BAP (0.5+1.0mg) and TLC study is also clear and distinct.
